This investigation was aimed to develop and evaluate lipid nanoemulsion-based gel of finasteride for topical administration, to increase drug availability into the skin for a long duration and to improve the in vivo potential of finasteride against alopecia. Nanoemulsion was prepared by a high-speed homogenization method using Vit E oil, cholesterol, and soya lecithin, characterized, and thickened using guar gum (nanoemulgel). Nanoemulgel was evaluated for pH, viscosity, drug release, skin permeability, and in vivo efficacy against alopecia. Size of the drug-loaded nanoemulsion droplets was observed to be 195.20 ± 9.43 nm with uniform size distribution (PDI; 0.25 ± 0.08) and negative zeta potential (− 7.61 ± 1.35 mV). The pH 5.37 ± 0.74 of the developed gel was found to be near to the skin pH. Gel showed slow release (94.77% in 24 h) and restricted permeation through the skin, i.e., 30.7% in 24 h. Macroscopic examination showed improved hair growth in the case of nanoemulgel. Hair diameter and length were observed to be increased significantly in the case of gel as compared with the testosterone treated group. The histological investigation supports the result of macroscopic examination where nanoemulgel restored the conditions (short length hair, necrotic cell appearance with miniaturized follicles) created by testosterone on the skin. Moreover, the developed formulation was observed to be safe and stable for at least 90 days. The results prove the clinical pertinence of this stable gel as it ensured longer stay over the skin, slower permeation and higher efficacy against alopecia.
Abstract Purpose We aimed to develop Squalene integrated NLC based carbopol 940 and HEC gel of tamoxifen citrate to increase lipid and moisture content of skin and to create a local drug depot in skin for improved efficacy against psoriasis. Methods: NLC was prepared by emulsification-cum-solidification method using high speed homogenizer, characterized and transformed to viscous gel having pH as similar as skin and smooth texture. Results and discussion: Developed gel showed the slow drug relea...

Background and Aim. Changes in total energy expenditure (TEE) and substrate metabolism may help explain the metabolic actions of testosterone (T). This study measured respiratory quotient (RQ), TEE, ghrelin, insulin, and key lipolysis enzyme concentrations in relation to body weight (wt) and food intake (FI) in both normal and bilaterally orchiectomized rats with/without T treatment. Methods. In total, thirty-two male Wistar rats (300–400 g) were divided into four groups (n = 8/group), including...

Abstract The aim of the investigation was to develop nanoemulsion (NE) based emulgel of Mentha essential oil (MEO: potent antifungal property) to ensure enriched and extended therapeutic effectiveness against various species of Candida and vaginal candidiasis. NE was prepared and optimized using central composite design. pH-triggered crosslinking of NE in carbopol 940 matrix (emulgel) was achieved by triethanolamine. Emulgel was evaluated for in vitro (against Candida spp.) and in vivo (efficacy...

Background: Androgenetic alopecia (AGA) is a common form of hair loss in Asian men. Although AGA is often regarded as a relatively minor dermatological condition, hair loss can impact self-image and is a main cause for anxiety and depression in some men. We have treated patients with AGA for seven years. Objective: The goal of this study was to evaluate the effectiveness of our combination therapy in Asian men with AGA. Participants: Between the years 2011 and 2017, 18,918 male patients were tre...

Human skin could be a prime target to deliver drugs into the human body as it is the largest organ of human body. However, the main challenge of delivering drug into the skin is the stratum corneum (SC), the outer layer of epidermis, which performs the main barrier function of the skin. Scientists have developed several techniques to overcome the barrier properties of the skin, which include other physical and chemical techniques. The most common and convenient technique is to use special formul...

The use of nanoemulsion in augmenting dermal and transdermal effectiveness of drugs has now well established. The development of nanoemulsion based semisolid dosage forms is an active area of present research. However, thickening or liquid-to-semisolid conversion of the nanoemulsions provides opportunities to the formulation scientist to explore novel means of solving instability issues during transformation. Extending knowledge about the explicit role of nature/magnitude of zeta potential, type...

Abstract Current work reports the development and optimization of clobitasol propionate (CP) and calcipotriol (CT) loaded nanoemulsion based gel for topical treatment of psoriasis. Components of nanoemulsion viz. , oil and surfactant/co-surfactant were selected depending upon solubility and emulsification potential respectively. The optimized ratio of 5:3:2 of Capmul MCM C8 EP, Cremophor RH 40 and Labrafil 1944 CS was selected. Carbopol 980 was used as gelling agent to achieve final drug concent...

Andrographolide (AP), a phytoconstituent of Andrographis paniculata is reported as a potent hepatoprotective agent. However, utility of this molecule is restricted due to its low aqueous solubility, gastric instability and hence low bioavailability. It was aimed to formulate and characterize AP-loaded, natural biopolymer stabilized, multilayered nano-hydrocolloid delivery system. Nanoemulsion (NE) was formulated using layer-by-layer (LbL) technology via electrostatic deposition of chitosan over ...

Background/Aims: Androgenetic alopecia is an extremely common dermatological disorder affecting both men and women. Oral finasteride (FNS), a synthetic 4-aza-3-oxosteroid compound with poor aqueous solubility, blocks the peripheral conversion of testosterone to dihydrotestosterone (DHT) in a significant reduction in DHT concentration, achieving satisfactory results in alopecia treatment. However, its oral intake generally causes severe side effects. Considering that there is currently no scienti...

Nanoemulgel is an emanating drug delivery system particularly being explored in research and development of various topical formulations for both pharmaceutical and cosmeceutical applications. Nanoemulgel consists of emulsion, which can be water in oil or oil in water type, incorporated into gel base. The gel base is prepared using gelling agent which modifies emulsion into non-greasy, more stable, and viscous formulation. Incorporation of hydrophobic drugs in a topical gel formulation along wit...

Introduction The current work aimed to formulate a novel chitosan-based finasteride nanosystem (FNS-NS) for skin delivery to optimize the drug availability in skin for a longer time and enhance ex vivo performance of finasteride against androgenic alopecia. Methods Both undecorated and chitosan decorated FNS-NSs were synthesized by a high energy emulsification technique. All the prepared nanosystems were further subjected to physicochemical characterizations like pH, viscosity, encapsulation eff...
