Abstract
Abstract Oral colon targeted mini-tablets containing 5-Fluorouracil (5-FU) were developed by using guar gum, pectin and Eudragit S100. Probiotics were co-administered to constantly replenish the gut microflora of colon that gets damaged due to pathophysiology of cancer and side effect of 5-FU. Replacement of microbial count ensures that the polysachharide used for sustaining the drug release gets digested to release the drug, once the formulation reaches the colon. The site-specific release of coated tablets was confirmed by dissolution studies in medium containing rat caecal contents (RCC), wherein, the formulation containing Eudragit S100, pectin and guar gum in the ratio of 04:03:03 w/w, showed less than 10% drug release in initial 5 h and immediate burst release (100%) between 5th to 10th hour. The cell line studies carried for pooled dissolution samples indicated that more than 90% cells were viable in all the cases till 5th hour. Significant (p

Following oral administration, gastric emptying is often a rate-limiting step in the absorption of drugs and is dependent on both physiological and pharmaceutical factors. To guide translation into humans, small animal imaging during pre-clinical studies has been increasingly used to localise the gastrointestinal transit of solid dosage forms. In contrast to humans, however, anaesthesia is usually required for effective imaging in animals which may have unintended effects on intestinal physiolog...

Abstract Fisetin is a plant derived flavonoid that possesses anti-cancer, anti-oxidant and anti-Parkinson's activities. However, due to its lipophilicity it suffers from dissolution rate limited oral bioavailability that reduces its therapeutic efficacy. In order to overcome this issue self-nanoemulsifying drug delivery system (SNEDDS) of fisetin were formulated and characterized for droplet size, shape, zeta potential, cell viability, dissolution and permeability studies. Prepared SNEDDS (1.2 m...

Abstract Modified Apple Polysaccharide (MAP) has been reported to cure colorectal diseases by up-regulating apoptosis and down regulating metastasis. In the present study, mesalamine (MES) and MAP mini tablets have been prepared and co-administered with probiotics to provide site specific release of drug. Probiotics along with MAP, which acts as a prebiotic would replenish the colonic microflora that have been compromised due to colorectal pathology. MES mini tablets were prepared keeping guar g...

Abstract In the present study, different solid self-nanoemulsifying drug delivery system (S-SNEDDS) was formulated by using porous hydrophilic and hydrophobic carriers to improve the dissolution rate and bioavailability of simvastatin (SIM). The prepared liquid-SNEDDS composed of Labrafil M 1944 CS/ Tween-80/ Ethanol (20% / 53.33%/26.67% v/v) with 0.1% SIM, resulted droplet size of 40.69 nm. The hydrophobic carriers used were Aerosil-200, Syloid 244FP, Syloid XDP 3150, Magnesium stearate, Micro ...

Abstract In the present study application of Modified Apple Polysaccharide (MAP) as tablet binder was evaluated. Liquid MAP was extracted from apple and solidified by adsorbing it on porous surface of Aerosil-200 and trehalose and this dispersion was dried using spray dryer. The concentration of excipients as well as spray drying conditions was optimised by using Box Behnken Design to achieve desirable powder characteristics. The optimised batch of solid MAP was characterized by DSC, PXRD, SEM, ...

OBJECTIVE: A major challenge in targeting orally administered drugs to colon is their passage through the long gastrointestinal path comprising highly variant conditions in terms of pH, viscosity, gut motility and microbial flora. Approaches to pH controlled release and microbially triggered release have proved to be successful in achieving colon targeting only to a partial extent. METHODS: In an attempt to improve targeting, both these approaches have been combined together with the approach of...

AbstractDevelopment of self-nanoemulsifying drug delivery systems (SNEDDS) of glimepiride is reported with the aim to achieve its oral delivery. Lauroglycol FCC, Tween-80, and ethanol were used as oil, surfactant, and co-surfactant, respectively as independent variables. The optimized composition of SNEDDS formulation (F1) was 10% v/v Lauroglycol FCC, 45% v/v Tween 80, 45% v/v ethanol, and 0.005% w/v glimepiride. Further, the optimized liquid SNEDDS were solidified through spray drying using var...

BACKGROUND: Polysaccharide based delivery systems have been successfully used to target drugs to colon. In some recent reports, the superiority of concomitant administration of probiotics with such systems has been established. However, the pharmacokinetics of such symbiotic therapy remain unexplored hitherto. METHODS: This study deciphers the pharmacokinetic parameters of guar gum based colon targeted spheroids of sulfasalazine with co-administration of probiotics in experimental rats. Thirty r...

The present study explains the effect of concomitant administration of synbiotics with the polysaccharide based colon targeted delivery system. As the gut microflora get deranged on administration of mesalamine, drug release from such delivery systems is expected to be jeopardised because the release trigger is dependent solely on the colonic microbiota. To overcome this limitation, mesalamine was formulated as microspheres with guar gum and xanthan gum (prebiotics). These were combined with pro...

AbstractThe aim of this study was to enhance the dissolution profile of the combination of glipizide and atorvastatin used for simultaneous treatment of hyperglycemia and hyperlipidemia. The strategy to formulate coamorphous glipizide–atorvastatin binary mixture was explored to achieve enhancement in dissolution. The coamorphous glipizide–atorvastatin mixtures (1:1, 1:2 and 2:1) were prepared by cryomilling and characterized with respect to their dissolution profiles, preformulation parameters a...

Abstract null null In case of colon-specific delivery of therapeutic agents through oral route, microbial/enzyme-triggered release approach has several advantages over other approaches due to unique microbial ecosystem in the colon. Multiple-unit carriers have an edge over single-unit carriers for this purpose. Among different materials/polymers explored, pectin appears as a promising biopolymer to construct microbial-triggered colon-specific carriers. Pectin specifically degrades by colonic enz...

Abstract The development of probiotic formulations has attracted great attention of researchers and industrial community. Their beneficial effects have been demonstrated in human health, and in buccal cavity including prevention of gingivitis, periodontitis, dental caries, oral candidiasis, and immunomodulation. The development of mucoadhesive wafers as buccal systems is important to promote intimate contact between the probiotics and oral mucosa, as well as maintain the viability of anaerobic b...
