Abstract Five new prenylated acetophenones, melicoptelins A-E (1–5), along with one known congener (6) were isolated from the roots of Melicope ptelefolia. Among them, compounds 2a/2b, 3a/3b, and 4a/4b were obtained as inseparable interconverting mixtures of keto and enol tautomers. Their structures were elucidated on the basis of extensive spectroscopic methods, including 1D, 2D NMR and HRESIMS. Compouds 2a/2b, 4a/4b and 5 exhibit protein tyrosine phosphatase 1B (PTP1B) inhibitory activity with IC50 values of 34.4, 55.2 and 66.6 μM, respectively.

Two polyprenylated acylcyclopentanone racemates, evodialones A (1) and B (2), featuring a 3-ethyl-1,1-diisopentyl-4-methylcyclopentane skeleton, were isolated from an extract of the aerial parts of Evodia lepta. Evodialone A (1) was resolved by chiral-phase HPLC to afford a pair of enantiomers, (+)- and (−)-evodialones A (1b/1a), while evodialone B (2) could not be resolved. Their structures were elucidated by spectroscopic analysis and a combination of computational techniques including gauge-i...

Nine dimeric acylphloroglucinols, including the new hyperjaponicols A–D (1–4), were isolated from the whole plant of Hypericum japonicum. The new structures were determined by the interpretation of NMR and MS data, and the relative configuration of the known compound, sarothralen C (5), was reassigned via NMR spectroscopic analysis and single-crystal X-ray diffraction data. The inhibitory activities of the isolates against four bacterial strains were evaluated, and compounds 1–4, 6, and 7 exhibi...

Four new acetophenone di-C-glycosides, pteleifolols A–D (1–4) and a new dimeric benzopyran, pteleifolol E (5), were isolated from the leaves of Melicope pteleifolia. Seven known compounds, including 2,4,6-trihydroxyacetophenone-3,5-di-C-glucopyranoside (6), were also isolated. Structures of the new compounds (1–5) were established by using spectroscopic and spectrometric techniques, including 1D and 2D nuclear magnetic resonance (NMR), UV, and high-resolution electrospray ionization mass spectro...

Abstract Bioactive substances that promote neurite outgrowth and prevent neuronal degeneration are potentially useful for the medical treatment of Alzheimer's disease (AD). This study aimed to obtain bioactive compounds from the stems of the medicinal and edible plant Melicope pteleifolia that promote neurite outgrowth. A bioassay-guided phytochemical investigation led to the isolation of four new chroman derivatives, pteleifolones A–D ( 1 – 4 ), and nine known components, acronyculatin B ( 5 ),...

Protein tyrosine phosphatase 1B (PTP1B) plays a vital role in the regulation of insulin sensitivity and dephosphorylation of the insulin receptor, so PTP1B inhibitors may be potential agents to treat type 2 diabetes. In this work, a series of novel imidazolidine-2,4-dione derivatives were designed, synthesized and assayed for their PTP1B inhibitory activities. These compounds exhibited potent activities with IC50 values at 0.57-172 μM. A 3D-QSAR study using CoMFA and CoMSIA techniques was carrie...

Melicolones A (1) and B (2), a pair of rearranged prenylated acetophenone epimers with an unusual 9-oxatricyclo[,8]nonane core, were isolated from the leaves of Melicope ptelefolia. Further chiral high-performance liquid chromatography resolution gave enantiomers (+)- and (−)-1, as well as (+)- and (−)-2, respectively. The structures and absolute configurations of the pure enantiomers were determined by extensive spectroscopic data and single crystal X-ray diffraction. All the isolated e...

Abstract Three new furoquinoline alkaloids, leptanoines A–C (1–3) along with three known compounds (4–6) were isolated from the leaves of Evodia lepta. Their structures were identified by interpretation of their spectroscopic data as well as comparison with those reported in the literature. In addition, all isolated compounds were evaluated for their acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) activities. Compound 4 showed the highest inhibitory activity towards BChE with an IC5...

The dichloromethane extract from leaves of Melicope barbigera (Rutaceae), endemic to the Hawaiian island of Kaua’i, yielded four new and three previously known acetophenones and 2H-chromenes, all found for the first time in M. barbigera. The structures of the new compounds obtained from the dichloromethane extract after purification by chromatographic methods were unambiguously elucidated by spectroscopic analyses including 1D/2D NMR spectroscopy and HRESIMS. The absolute configuration was deter...

Abstract The rhizome of Dryopteris crassirhizoma Nakai is an important traditional Chinese medicine listed in Chinese Pharmacopoeia for the treatment of parasitic infestation and viral diseases. In this work, ten enantiomeric pairs of new acylphloroglucinol meroterpenoids, (±)-dryocrassoids A − J [(±)-1−10], along with six known acylphloroglucinols (11−16) were isolated from the rhizomes of D. crassirhizoma. Notably, these new enantiomers were the first examples of meroterpenoids constructed by ...

Abstract Two new prenylated acetophenone derivatives racemates, meliviticines A ( 1 ) and B ( 2 ) with unprecedented rearranged skeletons, were isolated from Melicope viticina . Subsequent chiral resolution led to the separation of two pairs of enantiomers, (±)-meliviticines A ( 1a / 1b ) and (±)-meliviticines B ( 2a / 2b ). Their structures including absolute configurations were elucidated by extensive spectroscopic data, electronic circular dichroism analysis, and X-ray crystallography. A plau...
