Abstract
Michael-addition of a thiol to a maleimide is commonly used for bioconjugation of drugs to macromolecules. Indeed, both current FDA-approved antibody–drug conjugates—Brentuximab vedotin and Trastuzumab emtansine—and one approved PEGylated conjugate—Cimzia—contain a thiol–maleimide adduct.
