The goal of this investigation was to establish a reliable technique for the quantitation of ponazuril in limited sample volumes. Samples were extracted with chloroform and separation was achieved with a Symmetry RP18 column. Ultraviolet absorption was measured at 254 nm. A combination of 0.1% formic acid and acetonitrile (50:50) was used as the mobile phase. The calibration curve was linear from 0.1-25 μg/mL, with a lower limit of quantification of 0.1 μg/mL with a 100 μL sample. The precision ...

Chiral compounds commonly exist in traditional Chinese medicine (TCM) but little research on the quality control of TCM has been conducted. In this study, a new strategy is proposed, taking Yuanhuzhitong tablet (YHZT, consisting of Radix Angelicae Dahuricae and Rhizoma Corydalis, YHS) for example, which is based on Chiral isomers ratio analysis to monitor the production process of Chinese patent medicine (CPM) companies. In the process of content determination for tetrahydropalmatine (THP) in YH...

In this study, a new fluorinated methacrylamide chitosan (MACF) was synthesized and evaluated as adsorbent in the dispersive solid-phase extraction (d-SPE) for the effective determination and extraction of 20 organophosphorus pesticides from ginseng samples by the QuEChERS (Quick, Easy, Cheap, Effective, Rugged, Safe) method couple with gas chromatography-tandem mass spectrometry (GC-MS/MS). The properties of MACF were characterized by Fourier transform infrared spectroscopy, elemental analysis ...

In the management of cystic fibrosis, treatments against Staphylococcus aureus and Haemophilus influenzae such as amoxicillin or cotrimoxazole have to be prescribed and the antibiotherapy efficacy may be linked to the concentration that reaches the infected site. As cystic fibrosis patients present disturbed pharmacokinetics parameters, a drug monitoring would be relevant to assess lungs distribution of antibiotics and to optimize dosing regimens. In this context, the aim of the study was to dev...

Background and objectives null Dabrafenib is a novel small molecule tyrosine kinase inhibitor which used to treat metastatic melanoma. The aim of this research was to survey the effects of dabrafenib on human UDP-glucuronosyltransferases (UGTs) and to evaluate the risk of drug-drug interactions (DDIs). null Methods null The formation rates for 4-methylumbelliferone (4-MU) glucuronide and trifluoperazine-glucuronide in 12 recombinant human UGT isoforms with or without dabrafenib were measured, an...

Ticagrelor is a first-line clinical drug for the treatment of acute coronary syndrome, but its oral bioavailability is relatively low. Flavonoids (polyphenol compounds commonly found in plant foods) seriously affect human metabolism and health. This study compared the effects of quercetin, luteolin and catechin on the pharmacokinetic parameters of ticagrelor and found that quercetin can significantly increase the Cmax and AUC0-36 of ticagrelor, that is, quercetin can enhance the bioavailability ...

Monocrotaline (MCT) is a pyrrolizidine alkaloid that can induce hepatic sinusoidal damage, pulmonary hypertension, renal toxicity, and heart disease. Monocrotaline N-oxide (MNO), the primary metabolite of MCT, is less toxic; however, it can convert back to MCT to exhibit its toxicity. This study developed and validated a rapid and sensitive LC-MS/MS method for the simultaneous determination of MCT and monocrotaline N-oxide in rat plasma. The method has a linearity over the concentration range of...

To investigate the clinical pharmacokinetics of CA4P, a high-throughput high-performance liquid chromatography-tandem mass spectrometry assay with an identical positive electrospray ionization (ESI) mode was developed for the simultaneous determination of CA4P, its active metabolite CA4, and CA4 glucuronide in human plasma. CA4P and CA4 were easier to protonate in positive ESI mode, whereas CA4G was reported to produce deprotonated ion in negative ESI mode. Because the baseline separation of CA4...

The present study aimed to develop a rapid, sensitive, and specific LC-MS/MS method for the determination of nootkatone in rat plasma. α-Cyperone was chosen as the internal standard (IS). The plasma was processed using a one-step acetonitrile protein precipitation. Chromatographic separation of nootkatone was achieved on a Phenomenex Kinetex XB-C18 column (2.10 ×50 mm, 2.6μm) at 35 °C with a mobile phase consisting of acetonitrile and water under a gradient elution at a flow rate of 0.35mL/min. ...

Mammalian target of rapamycin (mTOR) drives its fundamental cellular functions through two distinct catalytic subunits, mTORC1 and mTORC2, and is frequently dysregulated in most cancers. To treat cancers, developed mTOR inhibitors have been classified into 1st - and 2nd -generations based on their ability to inhibit single (1st -generation) and dual (2nd -generation) mTOR subunits. However, the underlying metabolic differences due to the effects of 1st - and 2nd -generation mTOR inhibitors have ...
