Objective To investigate the effects of leukotriene C4 (LTC4) and Th1/Th2 imbalance in asthmatic guinea pigs and explore the mechanism of antiasthma by inhaled arsenic trioxide (As2O3) and moxa leaf oil.Methods The guinea pig asthma models were established with ovalbumin (OVA) challenge method.60 guinea pigs wen randomly divided into As2O3 atomization inhalation group (B1:4.0 mg/kg/d,B2:4.0 mg/kg/d),combined treatment group (C1:2.0 mg/kg/d + 0.05 ml,C2:2.0 mg/kg/d+0.1 ml),saline aerosol inhalation group (group A) and normal control group (group D).LTC4 and Th1/Th2 imbalance in bronchoalveolar lavage fluid (BALF) with all of the groups were compared after 7 days of inhalation.Results ① LTC4:compared with group A,B1,B2,C1,C2 and D groups were showing statistical significant difference (P ＜ 0.01); B1 and C1 groups showed statistical significant difference (P ＜ 0.01); B2 and C2 groups had statistical significant difference (P ＜ 0.05); B1 and B2 groups showed no statistical significant difference (P ＞ 0.05); ② Th1/Th2 imbalance:IFN-γ:compared with group A,B1,B2,C1,C2 and D groups were showing statistical significant difference (P ＜ 0.01); there were no statistical significant differences among the drug groups (P ＞ 0.05); IL-4:B1 and B2 groups had statistical significant difference (P ＜ 0.01); there were no statistical significant differences between B1 and C1 groups (P ＞ 0.05); there were no statistical significant difference between B2 and C2 groups (P ＞ 0.05) ; IFN-γ/IL-4:B1 and B2 groups were showing statistical significant difference (P ＜ 0.05); there were no statistical significant difference between B1 and C1 groups (P ＞ 0.05); there were no statistical significant difference between B2 and C2 groups (P ＞ 0.05).Conclusion The rise of LTC4 and Th1/Th2 imbalance were important pathogenesises of bronchial asthma.Arsenic trioxide could relieve asthma by reducing LTC4 and adjusting the imbalance of Th1/Th2.Moxa leaf oil could relieve asthma by reducing LTC4.The combination bv inhaled smaller doses of As2O3 and moxa leaf oil was safe and effective to relieve asthma.There were drug synergisms between As2O3 and moxa leaf oil.Key words: Arsenic trioxide;  Moxa leaf oil;  Aerosol inhalation;  Bronchial asthma;  Guinea pig;  Leukotriene C4;  Th 1/Th2 imbalance
