Abstract In this study, the hypoglycemic effect and possible mechanism of a polysaccharide, HEP-C, isolated from the fruit body of Hericium erinaceus were evaluated in streptozotoxin (STZ)-induced diabetic rats. Compared with the untreated STZ-induced diabetic rats, the supplements with HEP-C (150 and 300 mg/kg body weight [BW]) could significantly and dose-dependently relieve BW loss and organ injures, reduce fasting blood glucose, enhance glucose tolerance, alleviate hepatic function and serum lipid metabolism, elevate antioxidant enzyme activities, and suppress lipid peroxidation, which contributed to its potent hypoglycemic benefit. Liver histopathological observation revealed that HEP-C could effectively attenuate the deteriorated hepatic lesions in STZ-induced diabetic rats. HEP-C with potent hypoglycemic effect positively mediated glycogen synthesis by activating the phosphatidylinositol-3-kinase/protein kinase B signaling pathway. In summary, these results suggested that HEP-C, as a new dietary functional food or therapeutic agent, exhibited great potential for the prevention and treatment of diabetes mellitus and its complications.

Abstract Saponins as small organic molecules and polysaccharides as biomacromolecules are the main bioactive substances of Momordica charantia L. (M. charantia) with anti-hyperglycemic activities. This study was aimed to fully compare the antidiabetic effects and the potential mechanism of saponins (SMC) and polysaccharides (PMC) from M. charantia in STZ-induced type 2 diabetic mice with high-fat diet. Three dosages of SMC (L-SMC: 20 mg/kg, M-SMC: 40 mg/kg, H-SMC: 80 mg/kg) have a certain of the...

Here, the antioxidant and hypoglycemic activities of polysaccharides from Huidouba were preliminarily characterized. Two novel polysaccharides, HDBP-1 and HDBP-2 (127.33 kDa and 9.703 kDa, respectively), were purified by ultrafiltration using a Sephadex G-200 column and then characterized by high-performance gel permeation chromatography (HPGPC), high-performance liquid chromatography (HPLC), infrared spectroscopy (FT-IR), GC-MS, NMR and the Congo red test. HDBPs (crude Huidouba polysaccharide (...

Abstract In this study, hot water, 0.9% NaCl, citric acid, and 1.25 M NaOH/0.05% NaBH4 were separately used for the extraction of water-soluble H. erinaceus polysaccharides (HEPs; HEP-W, HEP-S, HEP-C, and HEP-A) from the fruit body of Hericium erinaceus. The physicochemical properties and biological activities were then investigated and compared. Results showed that the extraction solvents exhibited significant effects on the extraction yields, molecular weights, monosaccharide compositions, pre...

Abstract Three polysaccharides were extracted from Suillellus luridus (Suilu.A, Suilu.C and Suilu.S) by the hot water-extraction (1:40, w/v) and ethanol precipitation. Their preliminary structures were characterized and their antidiabetic activities were evaluated to obtain the most active component. The results of high performance gel permeation chromatography (HPGPC) and gas chromatograph (GC) showed that the main molecular weight of Suilu.A, Suilu.C and Suilu.S were 6383, 8172, 10,710 Da, and...

Mulberry fruit polysaccharide (MFP), one of the major active ingredients isolated from the mulberry fruit, possesses numerous bioactivities. In vitro hypoglycemic experiments showed that MFP had significant insulin-sensitizing activity through increasing insulin secretion and promoting pancreatic β cell proliferation. In vivo, the oral administration of MFP could significantly decrease blood glucose levels and increase body weight loss in streptozotocin (STZ)-induced diabetic mice. MFP also sign...
