Properties of soluble and structure-bound cyclic nucleotide phosphodiesterase were studied. No marked differences between these two forms of phosphodiesterase were found. The enzyme exhibits a pH-optimum at pH 8.0. Its activity is strongly dependent on Ca2+-concentration in the micromolar range. It is activated also by Mg2+-, Mn2+- and CO2+-ions and inhibited by Zn2+- and Cu2+-ions, methylxanthines, pyrophosphate, and ATP. The enzyme preferently hydrolyses 3′:5′-AMP. In the flight-muscles a heat-stable activator was detected. It was shown to be dialysable and resistant against enzymatic hydrolysis by trypsin and nagarse.
