Abstract This study aimed at investigating the synthesis, characterization, and search for a biotechnological application proposal for poly [(R)-3-hydroxybutyric acid] (PHB) grafted with the n-hydroxyethyl acrylamide (HEAA) monomer. The novel copolymer was prepared by 60Co gamma radiation-induced-graft polymerization. The effect of different solvents in the graft polymerization; the degree of grafting, crystallinity, and hydrophilicity; the morphology and the thermal properties were evaluated. The polyurethane fabricated from the grafted PHB was suggested as a scaffold. The enzymatic degradation behavior and the spectroscopic, morphological, mechanical, and biological properties of the composites were assessed. According to the results, the successful grafting of HEAA onto PHB was verified. The grafting was significantly affected by the type of solvent employed. A decreased crystallinity and increased hydrophilicity of the graft copolymer, concerning the PHB, was found. An increased roughness was observed in the morphology of the polymer after grafting. The thermodynamic parameters, except for the glass transition temperature, also decreased for the synthetic biopolymer. The intended use of these scaffolds for skin tissue engineering was supported by a proper degradability and degree of porosity, improved mechanical properties, the optimal culture of human fibroblasts, and its transfection with a plasmid vector containing an enhanced green fluorescent protein.

Abstract The synthesis of the poly(ester-urethanes) using poly(3-hydroxybutyrate) has gained increasing interest as scaffolds for use in tissue engineering despite their moderate hydrophobicity and biocompatibility. However, it is not yet known if the grafting of poly(N‐hydroxyethyl acrylamide) is suitable to improve the degree of hydrophilicity of the surface. This study aims to tailor the surface of these biopolymers by plasma-induced graft polymerization of N‐hydroxyethyl acrylamide. The synt...

Abstract Sucrose utilization has been established in Escherichia coli strains by expression of Mannheimia succiniciproducens β-fructofuranosidase (SacC), which hydrolyzes sucrose into glucose and fructose. Recombinant E. coli strains that can utilize sucrose were examined for their abilities to produce poly(3-hydroxybutyrate) [P(3HB)] and poly(3-hydroxybutyrate-co-lactate) [P(3HB-co-LA)] from sucrose. When recombinant E. coli strains expressing Ralstonia eutropha PhaCAB and SacC were cultured in...

The insufficient mechanical strength and slow degradation rate of poly-L-lactic acid (PLLA) limit its applications in bone repair. In this study, zeolitic imidazolate framework-8 (ZIF-8), one typical metal organic framework, was introduced into a laser-sintered PLLA scaffold. The results showed that the ZIF-8 formed favorable interface bonding with PLLA due to its inherent hydrophobicity and abundant organic ligands. Meanwhile, it served as an efficient nucleating site and increased the crystall...

Abstract The poor interfacial bonding and resultant agglomeration of nanoparticles in polymer-based composite severely deteriorated their reinforcement effect. In this work, MgO nanoparticles (MgO-NPs) were surface modified with Poly (L-lactic acid-co-malic acid) (PLMA) to improve the interfacial compatibility in Poly-l-lactic acid (PLLA) scaffold manufactured by selective laser sintering. PLMA possess a hydrophilic end with carboxyl group (comes from the malic acid) and an l-lactic acid chain. ...

Abstract Proper selection of materials and methods of construction are important factors in scaffolding in tissue engineering. In this research, a novel biomimetic scaffold containing poly (3-hydroxybutyrate) (PHB) and keratin (5-15wt. %) was prepared by the electrospinning method. The results showed that all electrospun scaffolds have 3D structures with high interconnected porosity and hydrophilicity. FTIR, Raman and H-NMR indicated the presence of keratin in electrospun scaffolds and the hydro...

: In recent years, naturally biodegradable polyhydroxyalkanoate (PHA) monopolymers have become focus of public attentions due to their good biocompatibility. However, due to its poor mechanical properties, high production costs, and limited functionality, its applications in materials, energy, and biomedical applications are greatly limited. In recent years, researchers have found that PHA copolymers have better thermal properties, mechanical processability, and physicochemical properties relati...

Abstract Poly-3-hydroxybutyrate (PHB) films were plasma-treated using pure NH3, pure Ar or mixtures of the two different proportions (20%, 30%, 40%, 50%, 70% NH3 in Ar). Surface chemistry and surface topography changes of PHB films were observed after plasma processing in all plasma regimes. The XPS results confirmed the absence of chemical modification in the case of pure Ar plasma treatment. Nitrogen-containing groups (e.g., N C O) were detected on the surfaces of P3HB films treated with NH3-c...

Abstract Reactive extrusion was used for dicumyl peroxide (DCP)-initiated grafting of glycidyl methacrylate (GMA) to poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV). The effects of GMA and DCP content and their weight ratio on the GMA grafting percentage (GP%), the polymer melt viscosity, and the PHBV molecular weight were investigated. FTIR spectroscopy determined that the DCP did indeed initiate GMA grafting. However, the changes in both the zero-shear viscosity ( η 0 ) and the molecular w...

Abstract A novel brush-like poly(2-aminoethyl methacrylate) (PAEMA) was grafted onto chitosan (CS) through gamma radiation-induced polymerization. The copolymer (CS-g-PAEMA) was used to prepare a sodium acetate leached poly(urethane-urea) scaffold. The above derivatives were developed, synthesized, and characterized to meet the specific characteristics of biomaterials. The results revealed that this method is an easy and successful route for grafting PAEMA onto CS. The feasibility of preparing a...
