Abstract Suzuki-Miyaura cross-coupling reaction of halogenated pyridines and a borated l -aspartic acid derivative was conducted. The reactivity of chloro-, bromo-, and iodo-pyridines with substituents at the C2, C3, and C4 positions was investigated. Electron density of halogenated pyridines was also estimated by density functional theory (DFT) calculations. Abstract An UV-light-induced S-methylation of quinoline-2-thione was completed by using dimethyl sulfoxide as methyl reagent. A series of quinoline methyl sulfides were prepared in moderate to good yields under environmentally friendly and mild conditions.

Abstract A series of novel 2-phosphonylated imidazo[1,2-a]pyridines was accessed from C N coupling of chloroethynylphosphonates and commercially available N-unsubstituted 2-aminopyridines. The product yield depends on the nature and position of the substituent in the pyridine ring.

A facile and complementary [3 + 2 + 1] annulation of aryl ketoxime acetates and 3-formylindoles to give pyridine derivatives is reported. The condensation reaction demonstrated that I2 was capable of triggering N–O bond cleavage of ketoxime acetates to generate iminyl radicals via a single electron transfer pathway. This direct and operationally simple protocol provides a fundamental platform to synthesize 3-(4-pyridyl)indoles with high functional group compatibility and high regioselectivity.

N-Methylated amino acids and peptides with an 8-aminoquinoline (AQ) directing group can be subjected to stereoselective Pd-catalyzed β-functionalizations. A highly efficient CoI2/o-phenanthroline catalyzed cycloaddition reaction of diynes bearing TBS protected propargylic alcohol fragments with nitriles has been developed. This methodology offers regioselective access, with good functional group tolerance, to various indeno[2,1-c]pyridine derivatives in moderate to excellent yields. It was found that o-phenanthroline as a bidentate nitrogen ligand showed high efficacy in this cycloaddition reaction.

Multicomponent reactions (MCRs) are robust tools for the rapid synthesis of complex, small molecule libraries for use in drug discovery and development. By utilizing MCR chemistry, we developed a protocol to functionalize the C-3 position of imidazo[1,2-a]pyridine through a three component, decarboxylation reaction involving imidazo[1,2-a]pyridine, glyoxalic acid, and boronic acid.
