Abstract Objective To explore the effect of acupuncture on the endoplasmic reticulum stress IRE 1-CHOP signal pathway and the expression levels of related apoptosis genes in pancreatic tissue of rats with STZ-induced diabetes mellitus. Methods Eighty SD rats were selected, among which, 10 were fed with normal diet and taken as blank control group (“blank group” for short), the other 70 rats were fed with high fat and high glucose food and injected with small dosage of STZ intraperitoneally, and 30 models were established successfully. The models were randomly divided into acupuncture treatment group (“acupuncture group” for short), model control group (“model group” for short) and western medicine group (metformin) with 10 rats in each group. After treatment for 4 weeks, RBG, and the expression levels of CHOP, IRE 1, Bax, Bcl-2 protein and mRNA in pancreatic tissue of rats were determined. Result Compared with the conditions before treatment, RBG reduced obviously in the acupuncture group and western medicine group ( P P >0.05); the expression levels of CHOP, IRE 1 and Bax proteins in pancreatic tissue of rats in the model group were significantly higher than those in the blank group ( P P P P P P P >0.05); the expression level of Bcl-2 mRNA in the model group was significantly lower than that in the blank group ( P P P >0.05). Conclusion There was endoplasmic reticulum stress in the pancreatic tissue of rats with STZ-induced diabetes mellitus, and IRE 1-CHOP signal pathway was the main pathway for inhibiting the endoplasmic reticulum stress in the pancreatic tissue. Acupuncture can improve the endoplasmic reticulum stress in pancreatic tissue, inhibit apoptosis genes, and protect the pancreatic tissue through IRE 1-CHOP signal pathway.

Abstract Exposure of Tca-8113 cells to proteasome inhibitor carbobenzoxy-Leu-Leu-leucinal (MG-132) causing apoptosis is associated with endoplasmic reticulum (ER) stress. X-box-binding protein-1 (XBP1) is an important regulator of a subset of genes active during ER stress, which is related to cell survival and is required for tumor growth. The present study is to evaluate the effect of MG-132 on ROS production, XBP1 gene expression, tumor necrosis factor receptor-associated factor 2 (TRAF2), ASK...

Pancreatic β cell failure leads to diabetes development. During disease progression, β cells adapt their secretory capacity to compensate the elevated glycaemia and the peripheral insulin resistance. This compensatory mechanism involves a fine-tuned regulation to modulate the endoplasmic reticulum (ER) capacity and quality control to prevent unfolded proinsulin accumulation, a major protein synthetized within the β cell. These signalling pathways are collectively termed unfolded protein response...

Cells exposed to ER-stress undergo the Unfolded Protein Response (UPR) to avoid apoptosis, but may also activate autophagy. However, the signal for selection of one of these two protective responses is unknown. To clarify the key switch between autophagy and apoptosis, we examined the correlation of UPR-related signals with autophagy and/or apoptosis inductions in HepG2 cells exposed to three ER-stress inducers (NaF, tunicamycin, and thapsigargin) with time, including the effect of small interfe...

The unfolded protein response (UPR) buffers protein-folding stress at the endoplasmic reticulum (ER). Research has provided insight into the regulatory mechanisms of UPR branches initiated by the stress sensors protein kinase RNA-like ER kinase (PERK), inositol-requiring protein 1α (IRE1α) and activating transcription factor 6 (ATF6), and into outcomes of the UPR that are not directly related to protein misfolding.

Endoplasmic reticulum (ER)-associated degradation (ERAD) is a mechanism against ER stress, wherein unfolded proteins accumulated in the ER are transported to the cytosol for degradation by the ubiquitin-proteasome system. We identiˆed the novel ubiquitin ligase HRD1 involved in ERAD. HRD1 is expressed in brain neurons and protects against ER stress-induced apoptosis. In familial Parkinson's disease, accumulation of Parkin-associated endothelin receptorlike receptor (Pael-R), a substrate of ubiqu...

Purpose: We have previously shown the prognostic significance of BCL2 expression in the activated B-cell–like diffuse large B-cell lymphoma (ABC-DLBCL) patients treated with cyclophosphamide-Adriamycin-vincristine-prednisone (CHOP) or CHOP-like therapy. However, after the inclusion of rituximab (R) in the CHOP regimen, several conflicting observations about the prognostic value of BCL2 expression have been reported. Experimental Design: We evaluated the R-CHOP cohort of 221 DLBCL cases with gene...

In pancreatic β-cells, the endoplasmic reticulum (ER) is an important cellular compartment for insulin biosynthesis, which accounts for half of the total protein production in these cells. Protein flux through the ER must be carefully monitored to prevent dysregulation of ER homeostasis and stress. ER stress elicits a signaling cascade known as the unfolded protein response (UPR), which influences both life and death decisions in cells. β-cell loss is a pathological component of both type 1 and ...

Changes in metabolic processes play a critical role in the survival or death of cells subjected to various stresses. In the present study, we have investigated the effects of ER (endoplasmic reticulum) stress on cellular metabolism. A major difficulty in studying metabolic responses to ER stress is that ER stress normally leads to apoptosis and metabolic changes observed in dying cells may be misleading. Therefore we have used IL-3 (interleukin 3)-dependent Bak −/− Bax −/− haemopoietic cells whi...

Inositol-requiring enzyme 1 (IRE1) is a proximal endoplasmic reticulum (ER) stress sensor and a central mediator of the unfolded protein response. In a human glioma model, inhibition of IRE1α correlated with down-regulation of prevalent proangiogenic factors such as VEGF-A, IL-1β, IL-6, and IL-8. Significant up-regulation of antiangiogenic gene transcripts was also apparent. These transcripts encode SPARC, decorin, thrombospondin-1, and other matrix proteins functionally linked to mesenchymal di...

Abstract IRE1, an ER-localized transmembrane protein, plays a central role in the unfolded protein response. Upon ER stress, IRE1 senses the accumulation of unfolded proteins in the ER, and transfers signal from the ER to the cytosol. Recently, it was reported that the luminal domain of yeast Ire1 senses the unfolded proteins via a two-step mechanism, namely dissociation of BiP and direct interaction with unfolded proteins. However, it has been unclear whether a similar mechanism is applicable t...
