Structure analysis jointly based on circular dichroism and Chou and Fasman analysis has been performed on a homologous group of 12 squash proteinase inhibitors. Using the solid-phase procedure, a linear 29-peptide with the reported sequence of trypsin inhibitor CMTI III was synthesized and oxidized to give three disulphide bridges. After affinity chromatography on an immobilized anhydrotrypsin column the synthetic product was shown by various physical techniques to be identical with the trypsin inhibitor CMTI III from squash seeds (Cucurbita maxima).

Abstract Seven new trypsin inhibitors, CyPTI I–VII, were purified from ripe seeds of Cyclanthera pedata by affinity chromatography on immobilized chymotrypsin in the presence of 5 M NaCl followed by preparative native PAGE at pH 8.9. A novel chymotrypsin inhibitor of the potato I protease inhibitor family from the earthworm Lumbricus terrestris was purified.
